TRIF also known as TICAM-1 is an adaptor protein containing a Toll/interleukin-1 receptor (TIR) homology domain, TRIF is an intracellular signaling domain that mediates protein-protein interactions between the Toll-like receptors (TLRs) and signal-transduction components. TRIF plays a crucial role in the immune response by mediating the signaling pathways activated by Toll-like receptor 3 (TLR3) in response to double-stranded RNA, which is a common viral signature. This interaction is vital for the production of interferon-beta, a key cytokine in antiviral defense, through the activation of transcription factors such as NF-kappaB, AP-1, and IRF3. The proper functioning of TIRF is essential for an effective immune response, and dysregulation can lead to impaired antiviral immunity or contribute to autoimmune diseases. |
