A20 also known as TNF alpha induced protein 3, TNFAIP3 is a critical Cys2/Cys2 zinc finger protein that plays a vital role in regulating inflammatory responses. A20 is primarily located in the cytoplasm and is induced by inflammatory stimuli such as tumor necrosis factor (TNF) and interleukin 1 (IL-1). A20 functions as a negative regulator of nuclear factor κ B (NFκB) signaling, which is essential for controlling immune responses and preventing excessive inflammation. By inhibiting NFκB activation, A20 helps terminate inflammatory signals, maintaining cellular homeostasis and preventing potential tissue damage. A20 structure, particularly seven zinc finger domains, is crucial for function, as only four domains are necessary for in vitro inhibition of TNF-induced NFκB activation. A20 interacts with several key proteins, including TRAF2, TRAF6, and IkB kinase (IKK) γ, further underscoring importance in cellular signaling pathways. |
