Flotillin-1 also known as FLOT1 is a member of the band 7.2/stomatin protein family, has a hydrophobic N-terminal region forming a transmembrane domain, integrating into the lipid bilayer. Flotillin-1 plays a crucial role in forming and maintaining lipid rafts, specialized membrane microdomains rich in sphingolipids and cholesterol, essential for signal transduction, membrane trafficking, and molecular sorting. These lipid rafts serve as platforms for assembling signaling complexes, influencing cellular responses to external stimuli. Notably, Flotillin-1 and Flotillin-2 expression levels are independently regulated, with Flotillin-2 being more ubiquitously expressed. Flotillin-1 association with estrogen receptor X (ER-X) facilitates rapid activation of the MAP kinase cascade, highlighting significance in cellular signaling pathways.
