EWS also known as EWSR1 is a multifunctional protein that is involved in various cellular processes, including gene expression, cell signaling, and RNA processing and transport. EWS plays a significant role in gene regulation and cellular processes. Notably, EWS gene is often involved in chromosomal translocations that lead to the formation of fusion proteins with transcription factors, such as ATF-1, which are implicated in human neoplasias. In Ewing sarcoma, EWS protein is fused to the DNA-binding domain of ETS transcription factors, including Fli-1, Erg, ETV1, E1AF, and FEV, resulting in the EWS/Fli-1 chimeric protein. This fusion protein acts as a potent transcriptional activator, enhancing the expression of target genes and promoting cell transformation, which is critical for tumorigenesis. |
