PDCD4(Programmed cell death 4) is a tumor suppressor and encodes a protein that binds to the eukaryotic translation initiation factor 4A1 and inhibits its function by preventing RNA binding. PDCD4 plays a crucial role in regulating apoptosis and functions as a tumor suppressor gene, particularly in lung cancer. PDCD4 inhibits the transformation of mouse keratinocytes induced by tumor promoters, highlighting its importance in cancer biology. PDCD4 is primarily located in the nucleus, where PDCD4 interacts with key regulatory proteins involved in gene expression and translation. Notably, PDCD4 directly binds to RNA helicase eIF4A, inhibiting protein synthesis by disrupting the formation of the cap-dependent translation initiation complex. This interaction underscores PDCD4′s significance in controlling cellular growth and proliferation, as PDCD4 also suppresses c-Jun activity, a critical transcription factor involved in oncogenic signaling pathways. |
