Bcl-10 also known as CIPER, c-CARMEN, mE10 was identified by its translocation in a case of mucosa-associated lymphoid tissue (MALT) lymphoma. Bcl-10 plays a crucial role in immune signaling and was initially identified as a gene that is truncated or mutated in MALT B cell lymphomas and other tumor types. Bcl-10 structure features an N-terminal caspase recruitment domain (CARD), which is essential for function in activating the NFκB signaling pathway via a NIK-dependent mechanism. This activation is significant as NFκB is a key regulator of immune responses and cell survival, making Bcl-10 a vital component in the modulation of apoptosis and inflammation. Furthermore, Bcl-10 has been implicated in the processing of caspase-9 to its active dimer, highlighting importance in apoptotic signaling. |
