CREB also known as CREB-1 is a crucial transcription factor that regulates gene expression by binding to the cAMP response element (CRE) in the promoters of target genes, thereby modulating essential cellular processes such as metabolism, neuronal plasticity, and survival. Upon phosphorylation at Serine 133 by protein kinase A, CREB-1 undergoes a conformational change that facilitates interaction with the CREB-binding protein (CBP), promoting the assembly of the basal transcription complex and activating transcriptional programs critical for cellular responses to hormonal and stress signals. This regulatory mechanism underscores CREB-1′s pivotal role in neoglucogenesis through interaction with the nuclear coactivator PGC-1, as well as involvement in the pathogenesis of type II diabetes and dilated cardiomyopathy, highlighting significance in both metabolic regulation and cardiovascular health. |
