ID3 also known as HEIR1 is a member of the Id family of basic helix-loop-helix (bHLH) proteins, ID3 plays a crucial role in cellular differentiation and proliferation by acting as a negative regulator of DNA binding for bHLH proteins. ID3 can dimerize with class A and B HLH proteins, preventing these complexes from binding to DNA and inhibiting gene expression necessary for muscle differentiation. As muscle cells differentiate, Id protein levels decrease, allowing for active heterodimer formation that promotes myogenic differentiation. Growth factors tightly regulate ID3 expression, and studies show reduced Id mRNA levels can delay quiescent cells′ reentry into the cell cycle upon growth factor stimulation, highlighting ID3 ′s significance in developmental processes and potential implications in muscle-related pathologies. |
