Notch3 is a critical member of the LIN-12/Notch family involved in cell fate determination, playing a pivotal role in regulating cell differentiation, proliferation, and apoptosis during development and in adult organisms. The Notch signaling pathway, mediated by interactions with ligands such as Jagged1, Jagged2, and Delta-like proteins, is essential for proper vascular development and function, particularly in vascular smooth muscle cells. Mutations in the Notch3 gene are associated with cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy (CADASIL), emphasizing its importance in vascular integrity and neurological health. Overexpression or rearrangement of Notch proteins, including Notch3, has been observed in various human tumors, indicating a role in oncogenesis and making Notch3 a potential target for cancer research. |
