LRP2 also known as GP330, Megalin is a member of the low-density lipoprotein receptor gene family, which includes other receptors like LDLR, LRP, VLDLR, and ApoER2. LRP2 is primarily located on the apical membrane of epithelial cells, particularly in the proximal kidney tubules, intestine, and ependymal cells, where LRP2 plays a crucial role in the reabsorption of essential molecules, including vitamin B12. Proper localization of LRP2 is vital for mediating the uptake of various ligands, such as apolipoprotein J (also known as Clusterin), which is associated with the clearance of beta-amyloid peptides linked to Alzheimer′s disease. Additionally, LRP2 serves as an antigenic determinant in Heymann nephritis in rats, highlighting megalin′s significance in kidney function and disease.
