K-Ras also known as c-Ki-ras is a crucial member of the Ras family of small GTPases, acting as a molecular switch in signal transduction pathways that regulate cell proliferation, differentiation, and survival. K-Ras functions as a sophisticated molecular switch in cellular signaling pathways through a carefully regulated cycle between active GTP-bound and inactive GDP-bound states. This cycling mechanism enables K-Ras to transmit signals from cell surface receptors to downstream effectors such as RAF kinases and PI3K, orchestrating fundamental cellular processes including proliferation, differentiation, and survival. The precision of this regulatory system proves essential for maintaining cellular homeostasis, as mutations at specific positions (particularly codons 12, 13, and 61) can lock K-Ras in its active state.
