HAND2 also known as dHAND is a member of the basic helix-loop-helix family of transcription factors. HAND2 plays a significant role in cardiac development, particularly in establishing left-right asymmetry in the heart. HAND2 is predominantly expressed on the right side of the looped heart tube and in the pulmonary ventricle, where HAND2 activates the transcription of essential genes such as Ufd1, which is involved in the ubiquitin-proteasome pathway, and Cdc45, a key player in DNA replication. Proper functioning of HAND2 is vital for the development of the heart′s structure, ensuring correct formation of heart chambers during cardiogenesis. HAND2 is expressed in sympathetic neurons and chromaffin cells throughout embryonic and fetal development, highlighting HAND2′s importance in neural crest development.
