TRAF6(TNF receptor associated factor 6) is a member of the TNF receptor associated factor (TRAF) protein family. TRAF plays a critical role in regulating adaptive immunity, innate immunity, and bone metabolism, making TRAF6 essential for maintaining immune homeostasis and skeletal integrity. As a ubiquitin ligase, TRAF6 is pivotal in mediating the activation of key protein kinases, such as transforming growth factor β-activated kinase (TAK1) and IκB kinase (IKK), through the formation of a unique polyubiquitin chain linked via Lys 63 of ubiquitin. This post-translational modification is crucial for the activation of the NFκB signaling pathway in response to interleukin-1 and Toll-like receptor ligands, highlighting TRAF6′s importance in immune signaling. The coiled-coil domain of TRAF6 is particularly significant for TRAF6 auto-ubiquitination and subsequent activation of the NFκB pathway. |
