SirT4 also known as SIR2L4, Sirtuin 4 is a member of the sirtuin family of proteins, homologs to the yeast Sir2 protein. SirT4 protein is primarily located in the mitochondrial matrix, where SirT4 is involved in the regulation of metabolic processes. The mitochondrial localization of SirT4 is significant because SirT4 directly influences mitochondrial function and energy metabolism, which are vital for maintaining cellular homeostasis. SirT4 has been shown to interact with insulin-degrading enzymes, and through ADP-ribosyltransferase activity, SirT4 negatively regulates insulin secretion from pancreatic β cells, highlighting SirT4′s importance in glucose metabolism and energy balance. The presence of a deacetylase sirtuin-type domain and the ability to bind zinc as a catalytic cofactor further underscore the functional versatility of SirT4 in cellular signaling pathways. |
