POLD1 also known as DNA pol δ, DNA polymerase delta 1 is composed of two main subunits, p125 and p50, with p125 directly interacting with the sliding DNA clamp protein PCNA, which enhances processivity during DNA synthesis. POLD1 plays a crucial role in DNA replication, recombination, and repair, which are essential processes for maintaining genome stability. POLD1 is particularly important for the synthesis of the lagging strand during DNA replication, where DNA pol δ processes Okazaki fragments, ensuring that the newly synthesized DNA is accurate and complete. POLD1 is involved in the repair of damaged DNA, excising erroneous or damaged segments to maintain genomic integrity. POLD1 is subject to regulation by various cell cycle proteins, highlighting POLD1′s importance in coordinating DNA synthesis with the cell cycle. |
