PML also known as TRIM19 is a member of the tripartite motif (TRIM) family. PML is a zinc finger transcription factor and is crucial for regulating gene expression involved in cell differentiation and apoptosis. PML′s role in these processes is particularly significant in acute promyelocytic leukemia, where PML gene undergoes a chromosomal translocation with the retinoic acid receptor α (RARα) gene, resulting in the formation of the PML/RARα chimeric protein. This aberrant protein disrupts normal myeloid differentiation by recruiting the nuclear co-repressor-histone deacetylase complex, which is essential for retinoic acid-dependent differentiation, and also induces gene hypermethylation, leading to the silencing of critical genes and promoting leukemogenesis.
