PTP1B also known as PTPN1 is the founding member of the protein tyrosine phosphatase (PTP) family, which was isolated and identified based on its enzymatic activity and amino acid sequence. PTP1B plays a crucial role in regulating insulin signaling and is involved in the negative regulation of the insulin receptor, making PTP1B a significant target for understanding metabolic disorders such as obesity and diabetes. Located primarily in the endoplasmic reticulum and cytoplasm, PTP1B′s positioning is essential for function, as PTP1B interacts with various signaling pathways that modulate cellular responses to insulin. PTP1B is synthesized as a 435 amino acid precursor, which is subsequently cleaved to produce the active 321 amino acid form, highlighting the importance of post-translational modifications in activation and function.
