ZO-2 also known as TJP2 is a zonula occluden that is a member of the membrane-associated guanylate kinase homolog family. ZO-2 plays a crucial role in forming and maintaining tight junctions, which are essential for regulating paracellular permeability and maintaining epithelial and endothelial barrier integrity. Tight junctions comprise various proteins, including ZO-2, ZO-1, and claudins, that work together to create a selective barrier controlling ion and molecule movement between cells. Proper functioning of ZO-2 is vital for tissue homeostasis and cellular signaling, as disruptions in tight junction integrity can lead to various pathologies, including cancer metastasis and inflammatory diseases. ZO-2 is specifically localized at tight junctions of epithelial cells, where ZO-2 interacts with other tight junction proteins and actin cytoskeleton, contributing to structural stability and dynamic regulation of these junctions. |
