TPX2(Targeting protein for Xklp2) is a crucial microtubule-associated protein that plays a significant role in the assembly of microtubules during cell division, particularly in the M phase of the cell cycle. TPX2 is activated by Ran-GTP, which facilitates chromatin-induced microtubule assembly, ensuring proper spindle formation and function. The structural integrity of TPX2 is vital as TPX2 interacts with Aurora-A kinase at the spindle apparatus, where TPX2 may be regulated through phosphorylation, further emphasizing TPX2′s importance in mitotic spindle assembly. Additionally, TPX2 activates Eg2, a kinase that is essential for microtubule dynamics, by enhancing Eg2′s phosphorylation and activity in a microtubule-dependent manner. Notably, TPX2′s function can be inhibited by binding to importin α, a nuclear import factor, which highlights the regulatory mechanisms that control TPX2′s activity. |
