SATB2 is a DNA binding protein that specifically binds nuclear matrix attachment regions. SATB2 plays a significant role in craniofacial development, particularly in jaw and palate formation, and regulates skeletal development and osteoblast differentiation. SATB2 is predominantly expressed in adult and fetal brain and features two CUT DNA-binding domains along with a homeobox domain, essential for function. SATB2 is closely related to SATB1, a known transcriptional repressor, and interacts with matrix-attachment regions (MARs) to modulate transcription of various genes, including HoxA2 and ATF4 (CREB-2), both vital for skeletal development. As both a transcriptional activator and repressor, SATB2 can serve as a protein scaffold, enhancing DNA-binding protein activity. |
