p40phox also known as NCF4 is a cytosolic regulatory component of the superoxide-producing phagocyte NADPH-oxidase, a multicomponent enzyme system important for host defense. p40phox plays a crucial role in activating the NADPH oxidase complex, essential for producing reactive oxygen species during immune response. p40phox is primarily located in the cytosol and translocates to the membrane upon neutrophil activation, where p40phox interacts with other cytosolic components such as p47phox and p67phox to form a functional oxidase complex. Proper localization and interaction of p40phox are vital for effective superoxide generation, critical for microbial killing and inflammation. p40phox undergoes post-translational modifications, including phosphorylation at specific sites, which regulate activity and interaction with other proteins in the NADPH oxidase complex. |
