Menin also known as MEN1, MEAI, SCG2 is a nuclear tumor suppressor that is mutated in patients with multiple endocrine neoplasia type I (MEN1). Menin plays a significant role in regulating gene transcription through covalent histone modifications, which are essential for cellular differentiation. Menin interacts with the TGFβ/Smad3 signaling pathway in osteoblasts, where Menin negatively regulates the transcriptional activities of BMP2/Smad1/5 and Runx2, thereby inhibiting late-stage differentiation. This regulatory function is vital for maintaining proper bone development and homeostasis. Menin influences the expression of insulin-like growth factor binding protein 2 (IGFBP-2) by modulating promoter activity, highlighting Menin′s role in growth factor signaling. Ectopic overexpression of Menin has been shown to induce apoptosis in murine embryonic fibroblasts in a Bax/Bak-dependent manner, further underscoring Menin′s importance in cellular regulation. |
