MetAP2 is a member of the methionyl aminopeptidase family. MetAP2 plays a crucial role in protein maturation by removing N-terminal methionine residues from nascent peptides, a process essential for proper folding and function of many proteins. MetAP-2 enzymatic activity regulates cell proliferation through various cellular processes that influence growth and differentiation. Neurofibromin (NF1) tightly regulates MetAP2 expression, and increased MetAP2 levels have been associated with several types of cancer, making MetAP2 a significant target for therapeutic intervention. MetAP2 inhibitors, such as ovalicin and fumagillin, have shown promise in cancer treatment by blocking MetAP2 activity, thereby potentially hindering tumor proliferation, especially in NF1-associated malignancies.
