FIH-1 also known as FIH, HIF1AN is a dioxygen-dependent asparaginyl hydroxylase that modifies target protein function by hydroxylating target protein asparagine residues. FIH-1 plays a crucial role in cellular response to hypoxia by regulating hypoxia-inducible factor 1 alpha (HIF-1α) activity. FIH-1 functions as an asparaginyl hydroxylase, catalyzing the hydroxylation of asparagine residue 803 within the carboxy-terminal transactivation domain of HIF-1α, which prevents interaction with co-activators and thus inhibits transcriptional activity. This regulatory mechanism is vital for maintaining cellular homeostasis under varying oxygen levels, as dysregulation of HIF-1α can lead to pathological conditions such as cancer and ischemia. |
