GFAT1 also known as GFPT1 is a member of the N-terminal nucleophile class of amidotransferases, converts fructose-6-phosphate into N-acetylglucosamine-6-phosphate. plays a crucial role as the first and rate-limiting enzyme in the hexosamine biosynthesis pathway, which is essential for the synthesis of glycoproteins and glycolipids that are vital for cellular functions. GFAT1 is primarily located in the cytoplasm, where GFAT1 catalyzes the conversion of fructose-6-phosphate into N-acetylglucosamine-6-phosphate, a precursor for the production of UDP-N-acetylglucosamine. This process links glucose metabolism to the regulation of cellular signaling pathways and gene expression, influencing various physiological processes. Dysregulation of GFAT1 activity has been implicated in metabolic disorders, including hyperglycemia-induced insulin resistance, which is a hallmark of type 2 diabetes.
