PFKM also known as PFK-1, PFKA plays a crucial role in glycolysis by catalyzing the conversion of fructose 6-phosphate and ATP into fructose 1,6-bisphosphate, a key regulatory step in glucose metabolism. This enzymatic activity maintains energy homeostasis in cells, particularly during periods of high energy demand. PFKM functions as a tetrameric enzyme composed of different isoforms, including muscle (PFKM), liver (PFKL), and platelet (PFKP) types, each regulated by various metabolites. ATP and citrate inhibit PFKM, signaling sufficient energy levels, while elevated AMP and fructose-2,6-bisphosphate activate PFKM, with fructose-2,6-bisphosphate being produced by the bifunctional enzyme PFK-2. This intricate metabolite regulation of PFKM underscores its importance in metabolic pathways and highlights potential therapeutic interventions in metabolic disorders.
