The mammalian Runt-related transcription factor (RUNX) family comprises three members, RUNX1, RUNX2 and RUNX3. RUNX family members are DNA-binding proteins that regulate the expression of genes involved in cellular differentiation and cell cycle progression. RUNX1 plays a critical role in hematopoiesis, the process of blood cell formation, and is essential for proper development of various blood cell lineages. RUNX1 regulates the expression of genes involved in cellular differentiation and cell cycle progression, making RUNX1 a key player in maintaining normal hematopoietic function. RUNX1 is frequently implicated in human leukemia due to chromosomal translocations that disrupt DNA-binding domain, leading to aberrant gene expression and oncogenesis. RUNX2 plays a critical role in skeletal development and mineralization, functioning as a transcription factor that regulates the expression of genes essential for osteoblast differentiation and bone formation. RUNX3 plays a crucial role in regulating gene expression related to cellular differentiation and cell cycle progression. RUNX3 is predominantly expressed in hematopoietic cells, including myeloid and B-cell lines, as well as in the spleen. |
