HDAC5(Histone deacetylase 5) is a member of the histone deacetylase family. HDAC5 is predominantly located in the nucleus but can shuttle between the nucleus and cytoplasm in response to cellular signals, a process regulated by phosphorylation and interaction with 14-3-3 proteins. This subcellular localization is crucial because, within the nucleus, HDAC5 deacetylates histone proteins, leading to a condensed chromatin structure and transcriptional repression of specific genes. Dynamic movement between cellular compartments allows HDAC5 to modulate gene expression, impacting processes like muscle differentiation, neuronal function, and stress responses. Dysregulation of HDAC5 localization or function has been implicated in various diseases, including cancer and cardiac hypertrophy, highlighting HDAC5′s importance as a therapeutic target.
