BLNK aslo known as SLP65 is a cytoplasmic linker or adaptor protein that plays a critical role in B cell development. BLNK is essential for B cell activation, proliferation, and differentiation. Located primarily in the cytoplasm, BLNK acts as a central hub for signaling pathways initiated by BCR engagement, facilitating the recruitment and activation of downstream effectors such as GRB2, Vav, Nck, and phospholipase C gamma (PLC γ). BLNK′s location in the cell enables signal integration from various kinases, including Syk and Lyn, which are vital for proper B cell development and function. BLNK exists in two isoforms, pp68 and pp70, resulting from alternative splicing of the human BLNK gene transcript, and phosphorylation by Syk is a key regulatory step that enables the activation of downstream signaling cascades. |
