PDGF-A also known as PDGF1 is a member of the protein family comprised of both platelet-derived growth factors (PDGF) and vascular endothelial growth factors (VEGF). PDGF-A protein plays a crucial role as a mitogen for mesenchyme- and glia-derived cells, consisting of two disulfide-bonded polypeptide chains, A and B, and existing as three isoforms: PDGF AA, PDGF AB, and PDGF BB. These isoforms exhibit varying affinities for two receptor types, α and β, which are structurally related and possess protein-tyrosine kinase domains. Binding of PDGF-A to receptors induces receptor dimerization and subsequent activation of receptor kinases, with A subunit specifically binding to α receptors with high affinity, while B subunit can interact with both α and β receptors.
