PI3 Kinase Class III also known PI3 kinase p100, VPS34; PIK3C3 is a catalytic subunit of the PI3K complex. PI3 Kinase Class III plays a crucial role in cellular processes such as endosome to lysosome transport, autophagy, and the formation of multivesicular bodies, which are essential for the degradation and recycling of cellular components. PI3 Kinase Class III is primarily located in the cytoplasm and is ubiquitously expressed, with significant levels found in skeletal muscle. PI3 Kinase Class III localization is vital for function in membrane trafficking and nutrient sensing, as PI3 Kinase Class III is involved in the phosphorylation of phosphatidylinositol to generate PtdIns3P, a lipid that serves as a signaling molecule in various cellular pathways. Notably, PI3 Kinase Class III operates differently from class I PI3Ks, as PI3 Kinase Class III activity is enhanced by manganese rather than magnesium, and PI3 Kinase Class III is subject to regulation by nutrient availability, highlighting PI3 Kinase Class III′s importance in the mTOR signaling pathway and autophagy regulation. |
