RPMI 1640 Medium is named after where it’s developed (Roswell Park Memorial Institute) and is a modification of McCoy’s 5A Medium. RPMI 1640 Medium was originally used to culture human leukemic cells in suspension cultivation and as a mono-layer. This medium was later found to be suitable for culturing a variety of mammalian cells including Hela, Jurkat, MCF-7, PC12, PBMC, astrocytes, and carcinomas.
RPMI 1640 consists glutathione and highly concentrated vitamins, which differs from other media. RPMI 1640 Medium contains biotin, Vitamin B12, PABA, highly concentrated inositol and choline chloride, which are not present in EMEM and DMEM.
RPMI 1640 Medium (ATCC modification) is modified based on the regular RPMI 1640 with an increase of glucose from 2.0 g/L to 4.5 g/L, as well as a decrease of sodium bicarbonate from 2.0 g/L to 1.5 g/L.
