T-bet also known as TBX21 is a member of a phylogenetically conserved family of genes that share a common DNA-binding domain, the T-box. T-bet plays a crucial role in the differentiation of T helper lymphocytes into Th1 cells, which are essential for immune response against intracellular pathogens. T-bet is a T box transcription factor that promotes expression of Th1 cytokine interferon-gamma (IFN-γ) while inhibiting development of Th2 cells, maintaining balance in immune response. This regulatory function helps prevent excessive Th2 responses that can lead to allergic reactions and asthma. T-bet is selectively expressed in Th1 cells, with expression upregulated by signals from T cell receptor and interleukin-12, highlighting importance in T cell activation and differentiation. |
