Rictor is a critical component of mTOR signaling pathway, which regulates cell growth, proliferation, and survival in response to nutrients and growth factors. Rictor′s ability to form a complex with FRAP is essential for integrating these signals, and Rictor plays a pivotal role in phosphorylation of Akt/PKB, a key player in cell survival and metabolism. Notably, Rictor directly phosphorylates Akt/PKB on Ser473, which is crucial for full activation, and Rictor also facilitates Thr308 phosphorylation by PDK1. This regulatory mechanism is particularly important in cancer context, as Rictor-FRAP complex may serve as a potential drug target in tumors that have lost expression of PTEN, a tumor suppressor that normally inhibits Akt/PKB activation. |
