GADD45α also known DDIT1, GADD45A is a member of a group of genes whose transcript levels are increased following stressful growth arrest conditions and treatment with DNA-damaging agents. GADD45α plays a crucial role in the cellular response to DNA damage, acting as a mediator between the p53 tumor suppressor pathway and DNA repair mechanisms. GADD45α interacts with cyclin-dependent kinases (Cdks) and proliferating cell nuclear antigen (PCNA), which are essential for cell cycle regulation and DNA replication. Through binding to these proteins, GADD45α inhibits Cdk activity, preventing cell cycle progression while enhancing DNA excision repair processes.
